About universal MosSCI sites

The universal MosSCI sites are insertion sites that allow insertion of the same transgene at several well-defined locations in the C. elegans genome. The method has the advantages:

  • One targeting vector is compatible with multiple insertion sites
  • The local genomic environment is the same for all insertions
  • Insertions can be followed by the adjacent marker (NeoR or GFP signal)

The method works by inserting a transgene into a well-defined landing site at specific locations in the genome. The insertion sites were engineered into the genome and are therefore all identical (except for variations in the co-insertion marker, NeoR or Pmyo-2:GFP:H2B). The insertion sites contain a Mos1 transposon that can be excised. After excision, a repair process copies the transgene from a targeting vector into the genomic location.

Requirements
  • The transgene cloned into a MosSCI targeting vector for the ttTi5605 site.
  • A strain containing the universal MosSCI landing site.
